DNA alternative created by scientists

Artificial genetic material – XNAs – expected to reveal how molecules first replicated and drive biotechnology research
Scientists have created artificial genetic material that can store information and evolve over generations in a similar way to DNA – a feat expected to drive research in medicine and biotechnology, and shed light on how molecules first replicated and assembled into life billions of years ago.
Ultimately, the creation of alternatives to DNA could enable scientists to make novel forms of life in the laboratory.
Researchers at the MRC Laboratory of Molecular Biology, in Cambridge, developed chemical procedures to turn DNA and RNA, the molecular blueprints for all known life, into six alternative genetic polymers called XNAs.
The process swaps the deoxyribose and ribose (the "d" and "r" in DNA and RNA) for other molecules. It was found the XNAs could form a double helix with DNA and were more stable than natural genetic material.
In the journal Science the researchers describe how they caused one of the XNAs to stick to a protein, an ability that might mean the polymers could be deployed as drugs working like antibodies.
Philipp Holliger, a senior author on the study, said the work proved that two hallmarks of life – heredity and evolution – were possible using alternatives to natural genetic material.
"There is nothing Goldilocks about DNA and RNA," Holliger told Science. "There is no overwhelming functional imperative for genetic systems or biology to be based on these two nucleic acids."
Vitor Pinheiro, a co-author on the paper, said the research could help scientists unpick how DNA and RNA became so crucial in the evolution of life, and perhaps even help in the search for extraterrestrial organisms. "If a genetic system doesn't have to be based on DNA and RNA, what then do you define as life? How do you look for life?" he said.
In an accompanying articleGerald Joyce, of the Scripps Research Institute in La Jolla, California, says the study heralds an "era of synthetic genetics, with implications for exobiology [which deals with extraterrestrial life], biotechnology and understanding life itself". He adds: "Construction of genetic systems based on alternative chemical platforms may ultimately lead to the synthesis of novel forms of life."
Other scientists, including a team at the J Craig Venter Institute, in Rockville, Maryland, are hoping to make synthetic organisms from scratch, but the majority of the work so far has used conventional DNA.
In his article on the Cambridge study Joyce alludes to the potential dangers of synthetic genetics. He writes: "As one contemplates all the alternative life forms that might be possible with XNAs and other more exotic genetic molecules, the words of Arthur C Clarke come to mind. In 2010: Odyssey Two, HAL the computer tells humanity, 'all these worlds are yours', but warns – 'except [Jupiter's moon] Europa, attempt no landings there'. Synthetic biologists are beginning to frolic on the worlds of alternative genetics but must not tread into areas that have the potential to harm our biology."

Physicists at the University of Dundee used equipment designed for
MRI-guided ultrasound surgery and created a tool that can lift and
spin a 10cm rubber disk with an ultrasound beam.

The team said the beam carries momentum that can push an object in its
path and can cause the object to rotate when shaped like a helix or
vortex.

Mike MacDonald, of the university's Institute for Medical Science and
Technology, said: "This experiment not only confirms a fundamental
physics theory but also demonstrates a new level of control over
ultrasound beams which can also be applied to non-invasive ultrasound
surgery, targeted drug delivery and ultrasonic manipulation of cells.

"The 'sonic screwdriver' device is also part of the EU-funded
Nanoporation project where we are already starting to push the
boundaries of what ultrasound can do in terms of targeted drug
delivery and targeted cellular surgery.

"It is an area that has great potential for developing new surgical
techniques, among other applications, something which Dundee is very
much at the forefront of."

Doctor Who uses his screwdriver to open locks and doors and control
other devices, but the Dundee team believes its study could help make
surgery using ultrasound techniques more precise and effective by
giving doctors the ability to steer ultrasound waves to the precise
spot where they are needed.

Ultrasound waves allow surgeons to treat a range of conditions without
having to cut a patient open.

Dr MacDonald added: "Like Doctor Who's own device, our sonic
screwdriver is capable of much more than just spinning things around."

The results of the research are published in the American Physical
Society's journal Physical Review Letters. Last year, scientists in
Bristol began working on their own "sonic screwdriver" to be used in
DIY. The team experimented with rotating ultrasonic force fields which
acted like the head of a screwdriver.

Miniature honeycomb 'grows nerve'

A "miniature honeycomb" - or scaffold - could one day be used to encourage damaged nerves to grow and recover, according to an international group of researchers.
The scaffold can channel clusters of nerves through its honeycomb of holes, eventually healing a severed nerve.
The findings of their study on mouse nerves are published in the journal Biofabrication.
Academics hope to one day treat spinal cord injuries with the scaffold.
When nerves are severed, such as in car accidents, it can result in a loss of feeling and movement.
Repairing this damage can be a challenge - but nerves outside of the brain and spinal cord can repair themselves, if only over short distances.

One technique to improve this repair is to use tubes. Either end of the severed nerve is placed in a tube and the two ends of the nerve should grow and join in the middle.
Researchers at the University of Sheffield and Laser Zentrum Hannover, Germany, investigated using a honeycomb structure.
Dr Frederik Claeyssens, from the department of materials science and engineering at Sheffield, told the BBC: "That is much more like the structure of the nerve itself.
"The nerve has small regions of 'cable' that go through from one end to the other end, you have a whole bunch of little cables inside a larger cable, that's what we tried to reproduce with this type of scaffold."
The honeycomb is made from photopolymerizable polylactic acid, which biodegrades once the nerve has repaired.
The researchers showed nerve cells could grow on the scaffold and are now testing it in mice to see if it can fully repair the damage.
Dr Frederik Claeyssens said: "This technology could make a huge difference to patients suffering severe nerve damage."
Scaffold technology is used in a range of "regenerative medicines". Building a scaffold and then coating it with human cells has, for example, been used to give patients new windpipes and bladders.

ON MARINE LE PEN AND POPULISM

Marine Le Pen and The French’s Front National are the big winners in the French elections yesterday. France’s Front National scored the best ever presidential campaign first-round result (18% of the votes).
As elsewhere in Europe, the French far right is dealing with matters other political parties prefer to avoid or shove under the carpet. Yesterday results proves that many French are primarily concerned with issues to do with immigration and ‘identity loss’. While the so called ‘far Right’ engages with these matters, the Left and the Centre parties perform an escapist attitude – they prefer to vet the discussion via different means such as political correctness and even legislation. The media, would also shy away from the subject and would prefer to gate-keep any attempt to deal with the ‘unpopular’ topic.
Le Pen’s victory is clearly an alarm call. As the financial turmoil starts to bite, it becomes clear that we are dealing with a ticking bomb. The way to defuse the situation is to launch a free and open discussion on maters to do with ‘belonging’, ‘identity’ and ‘culture’. The Left has been confused about it all for decades. European Left is riddled with contradiction, it would, for instance, support national movement around the world but never at home. The Left would adorably support Palestinian nationalism in Gaza and the West Bank but it would oppose similar English or French patriotism at home. How do we explain or justify such an unprincipled political attitude?
The Guardian refereed to Le Pen today as a ‘populist appeal’. It obviously missed the point once again. Le Pen is popular because she touches some (unpopular) issues no one else, (including the Guardian) dares to touch. It is in fact the Guardian and other media outlets that present a populist approach maintaining some delusional notions of correctness that appear to be detached from the reality in which we are living in.

The Crisis in Gaza

Have you heard much lately about the 1.5 million Palestinians illegally imprisoned by the Israeli government in the world’s largest open-air Gulag? Their dire living conditions, worsened by a selective Israeli siege limiting the importation of necessities of life – medical items, food, water, building materials, and fuel to list a few – has resulted in an 80 percent unemployment rate and widespread suffering from unlawful punishment, arbitrary arrests and imprisonment in Israeli jails.
The horrific conditions were a result of the Israeli invasion of Gaza in late 2008, ignited by Israel’s breaking of a truce with Gaza on November 4. Fourteen hundred people died, nearly three hundred of them children, and thousands were injured. The terror bombing of the Gazan population smashed into homes, hospitals, schools, ambulances, mosques, subsistence farms, UN facilities, and even the American International School. Israeli bombers destroyed over 30 members of one extended family in their home. That toll alone was three times the amount of Israeli fatalities, which included friendly fire.
The humanitarian crisis in crowded Gaza – about twice the size of the District of Columbia – “is now more dire than ever.” That is the judgment of Norwegian physician and professor of medicine, Dr. Mads Gilbert, who just finished a ten-day speaking tour in the U.S. Dr. Gilbert, returning from a recent visit to Gaza, was one of the only two foreign doctors inside Gaza during the massacre of December 2008 to January 2009.
He says: “During the Israeli attack, I saw the effects of new weapons including drones, phosphorous and also DIME [Dense Inert Metal Explosives], which leave no shrapnel, but I witnessed their capacity to cut a child in two; they also leave radioactive traces.”
Today, anemia and protein deficiency are widespread, reports Dr. Gilbert, especially among little children. UN and other relief supplies are inadequate, and UN humanitarian relief staff is often harassed by Israeli officials. Rebuilding pulverized Gaza is seriously obstructed by Israel blocking the imports of building materials.
Dr. Gilbert comments that he has “worked in other desperate situations in other places and Gaza is unique in a number of respects. It’s a captive population – usually if civilians are being attacked, there’s a safe place they can take refuge and then come
back to their homes when the fighting has stopped. That doesn’t exist for the people in Gaza since they are effectively imprisoned by the Israeli siege.”
Writing in the prestigious British medical journal “The Lancet” in early 2009, Dr. Gilbert provided clinical details of the slaughter, including the destruction of ambulances and medical facilities that tend to the dying and the wounded.
He described a “shattered, attacked, and drained health-care system trying to help an overwhelming amount of casualties in a war between clearly unequal powers, where the attacker spares no civilian lives – be it man, woman, or child – not even the much-needed health workers of all professions.”
It is no wonder the Israelis banned all foreign reporters, including those from the U.S. – the very country that provided the weaponry – thereby preventing the world from seeing the carnage as it happened.
The media ban made it possible for George W. Bush and president-elect Barack Obama to get away with describing this aggressive war with the identical phrase “Israel has the right to defend itself.” But apparently, the Palestinians do not have any way to defend themselves against the second-most modern military arsenal in the world; and their pleas about who broke the truce and started the bloodshed are unheeded.
Crude, garage-built Palestinian rockets are no match for modern precision missiles, helicopter gunships, bombers and drones. Fortunately for the Israelis, the rockets failed to reach any population centers 99 percent of the time. It was a mystery even to the Israelis why the unchallenged Israeli air force and ground artillery did not knock out the primitive Gaza launching sites, given its spies, informants and knowledge of every block in Gaza.
Reporters would have dug out these stories were they allowed inside Gaza. Since 2009, the focus of both the Israeli and U.S. government toward Iran has taken Gaza, the thousands of Palestinians in Israeli prisons, and the swallowing up of more land in the Palestinian West Bank, off of the news screens in the West.
It is remarkable how successful the Israeli propagandists have been in controlling the news coverage. They have even sidelined prominent retired Israeli security, military and political leaders, who along with civic and peace advocates are seeking a two-state solution, an end to confiscation of Palestinian land and houses, and debunking war talk against Iran, designed for domestic political purposes in Israel and the U.S.
For example, Meir Dagan, director of the Mossad – Israel’s CIA – from 2002 until 2010, called bombing Iran “the stupidest thing I ever heard.” In agreement are many other Israelis in the know. But, as in the U.S. during the months before the invasion of Iraq in March 2003, experienced voices of realism and sanity are not heard. Nor are sobering words of candor, as voiced by Israel’s founding father, David Ben-Gurion, who said, of the dispossessed Palestinians years ago, “we have come and we have stolen their country. Why would they accept that?”
Isn’t bringing these prominent Israeli truthsayers, peace advocates and military refuseniks to the U.S. Congress for their first-ever public hearing way overdue? At stake is peace or more wars in the Middle East. Also at stake is the possibility of another U.S. “war of choice” against Iran and the likely uncontrollable consequences that such belligerency would provoke. Would members of Congress let the AIPAC lobby block Israelis from coming here to present such testimony?
Or are the Senate Foreign Relations and House Foreign Affairs Committees, chaired respectively by Democratic Senator John Kerry and Republican Representative Ileana Ros-Lehtinen, satisfied with following their party lines?

record. That does not mean that there is any change to the Message which
says clearly that we must not use any drug except for medical purposes.
The same applies to the freedom of expression which must be absolute.
That does not mean again of course that I agree with anti-Jews,
anti-Semites, racists of any kind or anti-Raelians. But by knowing your
enemies or the enemies of your values, you are better equipped to fight
them. With love and respect of course, and with the wonderful sentence
of the French philosopher Voltaire in mind: "I disapprove of what you
say, but I will defend to the death your right to say it".

"Ethics" is simply a last-gasp attempt by deist conservatives and
orthodox dogmatics to keep humanity in ignorance and obscurantism,
through the well tried fermentation of fear, the fear of science and
new technologies.
 
There is nothing glorious about what our ancestors call history, 
it is simply a succession of mistakes, intolerances and violations.
 
On the contrary, let us embrace Science and the new technologies
unfettered, for it is these which will liberate mankind from the
myth of god, and free us from our age old fears, from disease,
death and the sweat of labour.

Food firms have been warned about the presence of a cancer-risk chemical in everyday products ranging from crisps and chips to instant coffee and ginger biscuits.

A biscuit designed for babies and toddlers has also been caught up in the alert.

Experts are even warning families to only lightly toast their bread at home, as the chemical, called acrylamide, is more likely to form the longer and darker foods cook.

A study by the Food Standards Agency has identified 13 products containing raised levels of the chemical. In each case, officials at the local council where the supplier is based have been told to notify them.

Acrylamide, which is still being investigated by scientists, is a cooking by-product associated with frying, baking, roasting or toasting foods at very high temperatures, usually greater than 120c.

The FSA insists its findings raise no immediate risk to the public and there is no need for people to change their diet.

However, it is putting pressure on all food companies to reduce acrylamide levels because long-term consumption could increase the risk of cancer. Its official advice is also that families should ensure bread and chips they eat are only toasted or baked to the 'lightest colour possible'.
 
The FSA said its study of levels of acrylamide and furan – another cancer-risk chemical – is used to identify which firms need to take action. Acrylamide is formed by a reaction between natural components in food as it cooks.

In reality it has probably been in the diet for as long as man has fried, roasted or toasted food. Manufacturers including Heinz and McVitie's have already responded by changing their recipes.

But others, including Nestle, makers of Nescafe, say it is impossible to do so without harming the flavour and quality of their products. It added: 'There is currently no scientific evidence to suggest any particular product has any negative impact on health in the context of acrylamide exposure.'

The FSA is required by the EU and the European Food Safety Authority to carry out the annual tests. It looked at 248 samples, from chips sold by fast-food outlets to supermarket own-label and big brand ranges. In 13 cases levels were above  the 'indicative value' – a trigger point to tell the firm it should examine its production process.

European watchdogs have been putting pressure on food manufacturers to reduce acrylamide for almost a decade. In 2002 Swedish studies revealed high levels formed during the frying or baking of potato or cereal products.

The FSA said: 'This raised worldwide public concern because studies in laboratory animals suggest acrylamide has the potential to cause cancer in humans by interacting with the DNA in cells.

'The Agency believes exposure to such chemicals should be as low as reasonably practicable.'

The latest survey found 'an upward trend' in acrylamide levels in processed cereal-based baby foods, excluding rusks. Importantly however, the FSA said this did not mean parents should stop giving these products to youngsters.

The Food and Drink Federation,  which represents manufacturers,  said members are 'ensuring levels are as low as reasonably achievable'.

Heinz changed its Banana Biscotti recipe this year to reduce acrylamide to trace levels. United Biscuits, which makes McVitie's Gingernuts, said it has cut acrylamide by 70 per cent. The firm also pledged to cut levels in its McCoy’s crisps.
