Scientists Develop Robot that Scans Information Directly from Neurons

Automated assistance may soon be available to neuroscientists tackling the brain’s complex circuitry, according to research presented last week at the Aspen Brain Forum in Colorado. Robots that can find and simultaneously record the activity of dozens of neurons in live animals could help researchers to reveal how connected cells interpret signals from one another and transmit information across brain areas — a task that would be impossible using single-neuron studies.
The robots are designed to perform whole-cell patch-clamping, a difficult but powerful method that allows neuroscientists to access neurons’ internal electrical workings, says Edward Boyden of the Massachusetts Institute of Technology in Cambridge, who is leading the work.
Manually performing the method on live animals requires extensive training to perfect and, as a result, only a handful of neurophysiologists use the technique, says Boyden, who presented at the conference. He is developing the automated tool with Craig Forest at the Georgia Institute of Technology in Atlanta and others. “We think that it helps democratize procedures that require a lot of skill,” he says.
In May, the group described how a basic version of the robot can record electrical currents in single neurons in the brains of anaesthetized mice. The robot finds its target on the basis of characteristic changes in the electrical environment near neurons. Then, the device nicks the cell’s membrane and seals itself around the tiny hole to access the neuron’s contents. On 24 August, Boyden presented results showing that a more advanced version of the robot could be used to identify and probe four neurons at once — and he says he wants to push the design further, perhaps to tap as many as 100 neurons at a time.
The researchers have also begun tweaking the robot to harvest the contents of cells, which could be tested for gene expression. Relating gene expression to neuronal activity could aid our understanding of what goes wrong in neurological disorders. “Maybe gene expression does specify electrical properties of neurons, and maybe it doesn’t. If it does, that would be fantastic to know,” says Boyden.

Neural recordings from live animals are most often collected using electrodes inserted between cells. These external recordings detect outgoing neuronal signals, but cannot measure the roiling electrical activity within each cell that determines whether and how rapidly a neuron fires.

It like it’s something out of a science-fiction movie – genius scientists engineer a synthetic skin that’s part living, part electronics.
But scientists at Harvard University have done just that, creating meshes of electronic and biological tissue.
The end result is cyborg tissue, which is created from electrodes and wires combined on a Nano-scale.

The results, published in Nature Materials, detail how scientists in the lab embedded electrical nanowires into the lab-grown flesh.
Dr Charles Lieber, who is a chemistry professor at Harvard and the leader of the research team, told the Harvard Gazette: ‘With this technology, for the first time, we can work at the same scale as the unit of biological system without interrupting it. 
‘Ultimately, this is about merging tissue with electronics in a way that it becomes difficult to determine where the tissue ends and the electronics begin.’
 
The Gazette notes that the researches initially worried about how the ‘skin,’ once implanted, would sense and react to chemical and electrical changes.
Normal human skin is capable of sensing oxygen, pH, and other elements in the air, and reacts to each one accordingly. 
The challenge, then, was engineering skin that would do the same.
First, a 3D mesh of organic polymer is laid out with nanoscale wires within. According to Nature Materials, the wires serve as ‘critical sensing elements.’

Then, they worked in human neurons, heart cells, and blood vessels.
When the substrate was dissolved, researchers had mesh they could contour into the shapes they needed.
Because of the embedded wiring, scientists were able to obtain accurate readings of pH.
Human cyborgs have been imagined in Hollywood for decades, famously in the Star Trek and Terminator franchises.
In both, the cyborg characters have decidedly human appearances, though below the epidermis still lurks a robotic core of metal.
However, the Harvard scientists are not looking to such lofty ends. 
Dr Lieber told the Gazette said their invention could greatly benefit the pharmaceutical industry, which could test its drugs on the cyborg skin instead of few layers of cultured cells.

Scientists have found that a man’s fertility could be restored by the growing of early stage sperm from a skin sample.
Research evidence suggests that adult cells, such as those of the skin, can be induced to return to a more primitive state and then turned into different cell types.
To see if it was possible to produce sperm cells, a team at the University of Pittsburgh School of Medicine in the US grew stem cells from skin samples and found they were able to generate key cells, including early stage sperm cells.
It is hoped the technique could help men who had cancer during childhood become fathers, as infertility can be a side effect of some cancer treatments.
Dr. Charles Easley, the study’s lead author, said: “Sperm can be banked for future artificial insemination procedures, but that does not help some patients, such as pre-pubertal boys.
“There are procedures to store testicular tissue prior to cancer therapy, but men who didn’t have the opportunity to save tissue are permanently sterile, and so far there are no cures for their sterility.”
The scientists say their findings show it might one day be possible to restore fertility for sterile men with an easily obtained skin sample.
“No one has been able to make human sperm from pluripotent stem cells (stem cells that can become almost any cell in the body) in the lab, but this research indicates it might be possible,” Dr. Easley said.
“This model also gives us a unique opportunity to study the molecular signals that govern the process, allowing us to learn much more about how sperm are made. “Perhaps one day this will lead to new ways of diagnosing and treating male infertility.”
The process is likely to raise concerns in some quarters however, as critics are wary of creating human life through artificial means and stem cell research remains controversial.
The latest development comes after British scientists claimed earlier this year to have made a major breakthrough by creating brain tissue from human skin.
